Application
Actigen is suitable for the preparation of anti-aging and anti-wrinkle skin care cosmetics.
Formulation : O/W emulsion The oil soluble part A is melted at 70°C. The water soluble part B is heated to 70°C, disperse xanthan gum into part B. Add part A into part B at 70°C and homogenize for 5 minutes. Cool down to 40°C and part C is added under vigorous mechanical stirring. Cool down to room temperature.

Test Methods : Actigen (Water/Glyceryl Polymethacrylate/Glycerin/3-aminopropyl dihydrogen phosphate)
Actigen is mixture of water, glyceryl polymethacrylate, glycerin and 3-aminopropyl dihydrogen phosphate. It contains 0.7~0.9 % of nitrogen (N : 14.01).
Appearance : Actigen is a colorless liquid having slightly characteristic odor
Identification
- Neutralize 5mL of Actigen with dilute sodium hydroxide TS, add silver nitrate TS: a yellow precipitate is formed. Then add dilute nitric acid or ammonia TS: the precipitate redissolves.
- To 0.5g of Actigen add 20mL of potassium hydroxide-ethanol TS, heat under a reflux condenser on a water bath for 2 hours. Cool, neutralize with dilute hydrochloric acid using phenolphthalein TS as an indicator, and evaporate this solution to make 20mL. Filter, if necessary, and heat the filtrate with 0.5g of potassium bisulfate: an acrolein-like, irritating odor is perceptible.
Purity
- Heavy metals Proceed with 1.0g of Actigen according to Method 2, and perform the test: the limit is not more than 20 ppm. Prepare the control solution with 2.0mL of Standard Lead Solution.
- Arsenic Take 1.0g of Actigen, prepare the test solution according to Method 3, and perform the test using Apparatus C with the test solution: the limit is not more than 2 ppm
Loss on drying : 87.0 ~ 90.0 % (1g, 105 , constant weight)
Nitrogen : Weigh accurately about 0.3g of Actigen and perform the test as directed under Nitrogen Determination (Method 1). Each mL of 0.01N sulfuric acid = 0.14007mg of N
Assay (3-aminopropyl dihydrogen phosphate)
- Preparation of sample solution
1.0 grams of Actigen was precisely weighed into 100ml volumetric flask, and purified water was added into the flask to make volume. The mixture was shaken vigorously. 5ml of this solution was taken into another 100ml flask, and the flask was filled with purified water to volume and shaken vigorously. This solution approximately contains 50ppm of 3-aminopropyl dihydrogen phosphate. Preparation of standard solutions 0.1 Grams of 3-aminopropyl dihydrogen phosphate standard sample was precisely weighed into 100ml volumetic flask, and purified water was added into the flask to make volume. After the mixture was shaken vigorously. 2.5ml, 5ml, and 10ml of this solution were taken into each 100ml flask, and the flasks were filled with purified water to volume and shaken vigorously. Each of these standard solutions contains 25, 50, 100ppm of 3-aminopropyl dihydrogen phosphate respectively.
- Assay methods
The concentration of 3-aminopropyl dihydrogen phosphate in ACTIGEN was determined by Ion Chromatography. The standard solutions were used to prepare the calibration curve, and the concentration of 3-aminopropyl dihydrogen phosphate in the sample solution was calculated from the calibration curve using peak area.
